| DNa Extraction | | 2008-07-14 16:09:00 | | DNA extraction is a routine procedure to collect DNA for subsequent molecular or forensic analysis. There are three basic steps in a DNA extraction:Breaking the cells open to expose the DNA within, such as by grinding or sonicating the sample.Removing membrane lipids by adding a detergent.Precipitating the DNA with an alcohol — usually ethanol or isopropanol. Since DNA is insoluble in these alcohols, it will aggregate together, giving a pellet on centrifugation. This step also removes alcohol-soluble salt.Refinements of the technique include adding a chelating agent to sequester divalent cations such as Mg2+ and Ca2+. This stops dnase enzymes from degrading the DNA.Cellular and histone proteins bound to the DNA can be removed prior to its precipitation either by adding a protease or having prior to precipitation, precipitating with sodium or ammonium acetate, or extracting with a phenol-chloroform mixture. If desired, the DNA can be resolubilized in a slightly alkaline buffer.Detecti | | By: Biosolution | | |

| TATA/Binding Protein DNA Complex | | 2008-06-23 08:31:00 | | TATA binding protein (TBP) is a transcription factor that binds specifically to a DNA sequence called the TATA box. This DNA sequence is found about 25-30 base pairs upstream of the transcription start site in some eukaryotic gene promoters. TBP, along with a variety of TBP-associated factors, make up the TFIID, a general transcription factor that in turn makes up part of the RNA polymerase II preinitiation complex. As one of the few proteins in the preinitation complex that binds DNA in a sequence-specific manner, it helps position RNA polymerase II over the transcription start site of the gene. However, it is estimated that only 10-20% of human promoters have TATA boxes. Therefore, TBP is probably not the only protein involved in positioning RNA polymerase II.
Role as Transcription Factor Subunit
TBP is a subunit of the eukaryotic transcription factor TFIID. TFIID is the first protein to bind to DNA during the formation of the pre-initiation transcription complex of RNA polyme | | By: Biosolution

| Paper: Mitochondrial DNA and maximum mammalian lifespan | | 2008-05-17 14:26:35 | | By Gilad Lehmann, Elena Segal, Khachik K. Muradian, and Vadim E. Fraifeld
In animal cells, mitochondria are semiautonomous organelles of virtually “hostile” (bacterial) origin, with their own code and genome. The semiautonomy and restricted resources could result in occasional “conflicts of interests” with other cellular components, in which mitochondria have greater chances to be “the weakest link,” thus limiting longevity. Two principal questions are addressed: (1) to what extent is mammalian maximum life span associated with mitochondrial DNA base composition? (2) Does mitochondrial DNA base composition correlate with another important mitochondria-associated variable -- resting metabolic rate -- and whether they complement each other in determination of mammalian maximum life span?

Keith Hirst was banged up in jail simply because he protested at how he was treated. He said yesterday he would fight to clear his name in a case which could leave him with a criminal record and cost taxpayers thousands of pounds. "The way I was treated you would have thought I had robbed a bank," he said. "My family are law-abiding people and I would help if I saw a gang of yobs attacking a police officer. This kind of incident does not help in improving relations between the community and police." I agree entirely.I cannot understand why the police seem totally oblivious to certain crimes and testosteronely crazy about others. | DNA chips and Microarrays | | 2008-04-23 21:04:00 | | A DNA microarray is an arrayed series of microscopic spots of DNA oligonucleotides, called features, where each feature contains picomoles of a specific unique sequence, such as a stretch of a gene sequence, which is used to measure the relative abundance of a sequence between samples, either differentially labelled or on different microarrays. These molecules are the used as probes to which only the correct target sequence will hybridise under high-stringency conditions; where the target is the labelled mix of either RNA or DNA to be analysed, in the majority of cases with a fluorophore-based detection system. In "traditional" microarrays the probes are bound to a solid surface by covalent attachment to a chemical matrix. The solid surface can either be glass or a silicon chip, in which case they are commonly known as gene, genome chip or colloquially Affy Chip when an Affymetrix chip is used. | DNA Replication Lecture | | 2008-04-10 22:22:00 | | DNA replication is the process of copying a double-stranded DNA molecule to form two double-stranded molecules.[1][2] The process of DNA replication is a fundamental process used by all living organisms as it is the basis for biological inheritance. As each DNA strand holds the same genetic information, both strands can serve as templates for the reproduction of the opposite strand. The template strand is preserved in its entirety and the new strand is assembled from nucleotides — this process is called "semiconservative replication". The resulting double-stranded DNA molecules are identical; proofreading and error-checking mechanisms exist to ensure near perfect fidelity.DNA Replication DNA Replication 2 Dna replication 3 In a cell, DNA replication must happen before cell division can occur. DNA synthesis begins at specific locations in the genome, called "origins", where the two strands of DNA are separated.[3] RNA primers attach to single stranded DNA and DNA polymerase extends fr | | By: Biosolution

| DNA Repair | | 2008-04-09 22:04:00 | | DNA repair refers to a collection of processes by which a cell identifies and corrects damage to the DNA molecules that encode its genome. In human cells, both normal metabolic activities and environmental factors such as UV light can cause DNA damage, resulting in as many as 1 million individual molecular lesions per cell per day. Many of these lesions cause structural damage to the DNA molecule and can alter or eliminate the cell's ability to transcribe the gene that the affected DNA encodes. Other lesions induce potentially harmful mutations in the cell's genome, which affect the survival of its daughter cells after it undergoes mitosis. Consequently, the DNA repair process is constantly active as it responds to damage in the DNA structure. The rate of DNA repair is dependent on many factors, including the cell type, the age of the cell, and the extracellular environment. | Can DNA & cloning return Morocco's lions to the wild? | | 2008-03-29 04:51:00 | | "The last known Barbary Lion in the wild was shot in 1942 on the northern side of the Tizi-n-Tichka pass in the Atlas Mountains, near the road between Marrakech and Ouarzazat, two of major tourists destinations in Morocco today."Two medieval skulls found in the Tower of London belonged to a kind of lion that boasted a giant dark mane, according to a genetic study that sheds new light on one of the world's oldest zoos. Infamous as a place of torture and executions, and home to the Crown Jewels, the Tower was also home to lions, which were charismatic symbols of monarchy. Now researchers have used DNA evidence to analyse two members of the royal menagerie, the oldest being late 13th to late 14th century (1280-1385) and 'youngest' 15th century (1420-1480), the only medieval big cat remains found in England. They conclude that they were male Barbary lions, a species that hails from north Africa, where no natural lion population remains today. | DNA sequencing | | 2008-03-28 21:44:00 | | DNA sequencing encompasses biochemical methods for determining the order of the nucleotide bases, adenine, guanine, cytosine, and thymine, in a DNA oligonucleotide. The sequence of DNA constitutes the heritable genetic information in nuclei, plasmids, mitochondria, and chloroplasts that forms the basis for the developmental programs of all living organisms. Determining the DNA sequence is therefore useful in basic research studying fundamental biological processes, as well as in applied fields such as diagnostic or forensic research. The advent of DNA sequencing has significantly accelerated biological research and discovery. The rapid speed of sequencing attainable with modern DNA sequencing technology has been instrumental in the large-scale sequencing of the human genome, in the Human Genome Project. Related projects, often by scientific collaboration across continents, have generated the complete DNA sequences of many animal, plant, and microbial genome.chain-termination or Sanger | | By: Biosolution | | |

| DNA Sequencers | | 2008-03-26 01:38:00 | | DNA sequencers have become more important due to large genomics projects and the need to increase productivity.Modern automated DNA sequencing instruments (called DNA sequencers) are able to sequence as many as 384 fluoresecently labeled samples in a batch (run) and perform as many as 24 runs a day. These perform only the size separation and peak reading; the actual sequencing reaction(s), cleanup and resuspension in a suitable buffer must be performed separately. The magnitude of the fluorescent signal is related to the number of strands of DNA that are in the reaction. If the initial amount of DNA is small, the signals will be weak. However, the properties of PCR allow one to increase the signal by increasing the number of cycles in the PCR program.A simple DNA sequencer will have one or more lasers that emit at a wavelength that is absorbed by the fluorescent dye that has been attached to the DNA strand of interest. It will then have one or more optical detectors that can detect at t | | By: Biosolution | | |

| Scientists find ET Genes in Human DNA. | | 2007-09-03 21:00:33 | | A group of researchers working at the Human Genome Project indicate that they made an astonishing scientific discovery: They believe so-called 97% non-coding sequences in human DNA is no less than genetic code of extraterrestrial life forms. The non-coding sequences are common to all living organisms on Earth, from moulds to fish to humans. In human DNA, they constitute larger part of the total genome, says Prof. Sam Chang, the group leader. Non-coding sequences, originally known as "junk DNA", were discovered years ago, and their function remained a mystery. The overwhelming majority of Human DNA is “Off-world” in origin. The apparent “extraterrestrial junk genes” merely “enjoy the ride” with hard working active genes, passed from generation to generation more

| Oldest DNA Ever Recovered Shows Warmer Planet | | 2007-07-08 12:20:00 | | DNA recently found shows that Greenland was once an area with trees and insects thriving on it.This is another example in the huge list of information that suggests that our planet has gone through trends of warming and cooling over the centuries. Scientists are now suggesting that perhaps trends in the sun's electro-magnetic radiation may be the cause of our temperature changes, and not any emissions we're putting out.Nowadays, the global warming debate has really turned more into a personal belief situation. You can pretty much find data to support either theory, and data to poke holes in that information as well. The big question isn't does it exist or is it something else, it's what we can do. As I've said many times, let's implement those new technologies in our own homes. Let's use less energy and reduce our dependency on foriegn oil. By doing these simple things we cannot only decrease emissions, but we can also save ourselves money in the long run. That is the pulse | | By: The Dissident Awakens | | |

| Soda Damages DNA | | 2007-05-30 16:02:00 | | Sodium benzoate, a preservative used in large quantities in soft drinks like Sprite, Dr. Pepper and Oasis, may cause severe damage to DNA and increase your risk of related diseases like Parkinson's while accelerating the aging process.Sodium benzoate appears to damage the “power station” of cells, known as the mitochondria, according to a British aging expert at Sheffield University. His studies found that the preservative can “totally inactivate” and “knock out” cells’ mitochondria, which causes the cells to seriously malfunction. The end result is an increased risk of a slew of diseases.Sodium benzoate has also come under fire because, when combined with the vitamin C in many soft drinks, it forms benzene, a cancer-causing chemical linked to leukemia. In one study, at least five brands of soft drinks were found to contain excessive benzene levels.Sodium benzoate is used freely in the United States and the UK. | DNA Security System | | 2007-05-24 17:06:20 | |
What about putting some DNA on your wife/husband so everyone know he/she is yours.
We are now able to provide you with a true DNA security marking system which holds Secured By Design accreditation. For the latest in marking and identifying your property SelectaDNA is a unique theft deterrent which really works.
Used in conjunction with the warning and window stickers supplied, the DNA marking system will help prevent the theft of all the expensive items in your home or work place. SelectaDNA comes in a pot of adhesive glue to apply to any valuable items of property. Once applied, removal of all the DNA marking from an item is almost impossible, as it is difficult to locate (except with a UV lamp) and remnants of the DNA will easily remain in crevices and uneven surfaces.
The DNA code along with the customers details are held on our secure LPCB approved database thereby allowing recovered property to be traced back to its rightful owner after analysis - even a tiny molecule of glue | | By: Random Good Stuff

| Hippocratus, Alcohol, Tobacco, DNA Aging, Chemotherapy and Cancer | | 2007-02-19 09:09:00 | | Hippocrates described several kinds of cancers. He called benign tumours oncos, Greek for swelling, and malignant tumours carcinos, Greek for crab or crayfish. The most consistent finding, over decades of research, is the strong association between tobacco use and cancers of many sites. Hundreds of epidemiological studies have confirmed this association. Lifestyle choices cause cancer: tobacco, diet, exercise, sex, alcohol, and tanning choices are the major risks. Most cancers are related to known lifestyle factors.Cancer can be treated by surgery, chemotherapy, radiation therapy, immunotherapy, monoclonal antibody therapy or other methods. The choice of therapy depends upon the location and grade of the tumor and the stage of the disease, as well as the general state of the patient (performance status). A number of experimental cancer treatments are also under development. Complete removal of the cancer without damage to the rest of the body is the goal of treatment.Much of the promise | | By: The Health Archive | | |
